Finally, Aquarius enables the dissociation of PRP2, plus the SF3A and SF3B complexes, which promotes the relocation of the branch duplex for catalysis. Translocation terminates six nucleotides downstream of the branch site through an assembly of PPIL4, SKIP and the amino-terminal domain of PRP2. Notably, PRP2 translocates along the intron while it strips away the RES complex, opens the SF3B1 clamp and unfastens the branch helix. The cryogenic electron microscopy structure of B AQR reveals how PRP2 and Aquarius remodel B act and B AQR, respectively. Here the inactivation of Aquarius leads to the stalling of a spliceosome intermediate-the B AQR complex-found halfway through the catalytic activation process. The role of Aquarius in splicing has been enigmatic 6, 7. ![]() Here we show that catalytic activation occurs in two ATP-dependent stages driven by two helicases: PRP2 and Aquarius. However, because PRP2 is observed only at the periphery of spliceosomes 3, 4, 5, its function has remained elusive. Catalytic activation occurs through an ATP-dependent remodelling mediated by the helicase PRP2 (also known as DHX16) 1, 2, 3. A crucial event of the splicing pathway is the catalytic activation, which takes place at the transition between the activated B act and the branching-competent B * spliceosomes. Pre-mRNA splicing follows a pathway driven by ATP-dependent RNA helicases. I already knew much of what I read in the book, but it answered a lot of questions I had and explained things better.) However, Nintendo has been traditionally greedy to a fault (it is what allowed Sony to unseat the company from its throne in the mid 1990s), and they know that they hold all of the leverage here. The point is, Nintendo could have kept Tears of the Kingdom priced at $59.99 and they would have made a mountain of profit on the title, regardless of the cost of its development. Nintendo doesn’t discount its Switch software, and it only discounted BoW this month ($39.99) in order to get players who haven’t played it yet invested on the franchise before Tears of the Kingdom arrives on May 12. What’s most impressive about Breath of the Wild, commercially speaking, is that it sold all of those copies at $59.99. Given that sales tend to slow down with the passage of time, and that FromSoftware does not really tend update their games, it will be hard for Elden Ring to reach those Breath of the Wild numbers, even over the next four years. Only Elden Ring, a title available on multiple systems, has been able to achieve similar success with 17 million copies sold by November 2022. That’s a staggering number copies for a Japanese Action-RPG that saw limited copies released on the Wii U, basically making it a single platform title on the Nintendo Switch. Sure, the development of a AAA Zelda game has always been a costly affair, but 2017’s Breath of the Wild had sold over 30 million units by April 2022. An engine made for a 2012 console which was already way behind the times (in terms of tech) 10 years ago. Well, the newest Legend of Zelda, a game slated for a May 12 release on Nintendo’s Switch, was built on Breath of the Wild’s engine. So, by now most of us are accustomed to paying $70 dollars for “next”, or should I say “current”, generation titles. The Legend of Zelda: Tears of the Kingdom is Announced to Have a Tear Inducing Price of $70 dollars Otherwise, Square Enix is in for a rough 2023. It might be time for Square Enix’s executives to get on their knees and engage in faithful prayer hoping that Final Fantasy XVI turns out to be a masterpiece at launch. Square Enix has already moved its biggest projects to Unreal 4 and it is likely that in the future the company will just move to the Unreal 5 engine, as well. Forspoken is a PS5 only game, and yet, it manages to look worse in spots than the aforementioned Final Fantasy XV did years ago on less powerful hardware. However, difficulties in the development of Final Fantasy XV, and now, a less than stellar performance (with some truly puzzling visual hiccups) in Forspoken will likely spell the end for the engine. This engine was once (in its early days) touted as a possible competitor to the Unreal 4 Engine. Square Enix has failed to provide any numbers for the game, as of the date of the publishing of this article, on Western territories, which is always a red flag.įorspoken’s critical, and (likely) commercial failure might have signaled the end of Square Enix’s use of its proprietary Luminous Engine. ![]() ![]() Japan’s physical sales of the game were pedestrian at 23,000. Scores like this, more often than not, translate into poor sales. Forspoken arrived to heavy criticism of its visuals, storyline and over all design, earning a terrible 65 Metacritic average, and an even lower 3.1 User average on the same site. New IPs that turn into bad games full of technical hiccups are doomed propositions. While I have grown less fond of Sonic by the passing year, my 6 year old kid is obsessed with anything Sonic related, and it is thanks to that obsession that I have been playing Sonic Frontiers with him. Kids (and even a segment of the adult gaming population) love Sonic. Still, when Sega designed the cute blue hedgehog, and its supporting characters in the late 1980s and 1990s, the company had stumbled upon a commercial winner. ![]() Sonic Has a Bright Future…I ThinkĮver since Sonic Adventure 2, it could be argued that Sonic 3-D games have been pedestrian at best, and completely terrible at worst. Out of the three headlines that we will discuss here, only one is positive, and that’s Sonic Frontiers’ commercial success despite the game’s ho-hum critical response of 71 Metacritic (C’mon the game is far from a masterpiece, but it is the best Sonic in decades, perhaps a low 80s rating was warranted here). One would think that we might have travelled back to the 1990s, but sadly, that is not case. Square Enix, Sega and Nintendo, have all made head lines this week. ![]() ![]() One important benefit of outliners is that there's absolutely no need to organize content in folders and fiddle with files and renaming stuff you just continually edit your "infinite" outline. Interestingly, DynaList has been created by the same Indie Hackers behind Obsidian. You can also "zoom" into a bullet point as if it was a document, and then you only see what's inside, meaning that you can actually store A TON of information. LogSeq / DynaList: Those are infinite outliners, which is a different style for note-taking. ![]() I love using it as a tool to think, reflect and explore ideas. You can also edit multiple documents at once, which is super useful. Obsidian has very good support for linking ideas, tagging and searching. You can publish your content to create a so-called "Digital Garden" basically exporting your notes as a static Website you can host anywhere. You can tweak it in lots of ways (not as many as emacs, but still □).There's a huge & friendly community around.Everything is stored locally in a single folder (Markdown files) that you can easily synchronize (e.g., using Google Drive, OneDrive etc) across devices.Obsidian: I currently use it for most of my knowledge management.I would not recommend Notion for personal knowledge management (assuming you write a lot and want to benefit from your writing in some way vs just keeping a journal from time to time). The versatility of the blocks is mind blowing you can do a ton of different things with Notion thanks to those. Notion: I use it as a place to store information about my projects, my home, my family, my processes, to maintain databases of different kinds (eg personal CRM).I've spent a good chunk of 2021 researching the note-taking and Personal Knowledge Management space for my project. But it's more about novelty than convenience, so I won't go into more depth about it. If I'm away from my computer and I want to take notes, I'll pop Dynalist open on my smartphone (it's right there in the dock for easy access) and dash off a few of the key points in my head.īonus: I also occasionally take notes on a ReMarkable e-ink tablet late at night when I want to avoid screens. In short, if I'm already at my computer working and I want to take notes, I'll do so in Notion. (If you're familiar with Workflowy, Dynalist is basically the same thing but with a few more features.) So I supplement Notion with a second app: Dynalist. And sometimes I need to be able to just jot a thought down quickly while cooking food or walking outside. However, Notion is slow af and has a recent history of crashing entirely. I'm a huge fan of Notion, and I take most of my notes there so that my notes can "live" in the same app that I also do the rest of my work - including scheduling tasks, tracking progress, managing my CRM, and more. It's more to do with getting as many songs as we could effectively within a budget. Were there any tracks you wanted but couldn't have?ĪC: Because it's mature, we're not gratuitous in that sort of way, there wasn't an issue of people wouldn't want to be associated because it was very violent. I'm not sure how many tracks there are - maybe 130? Definitely over a hundred. They have all this 50s music, and then there's the 40s music as well -being able to put all this big list together has been pretty cool for the guys. I'm not a big music guy myself, but obviously other guys on the team are, and for them it's like being kids in a candy store. But the music is obviously a big part of creating the period. carte blanche isn't true, it's not like we had billions of dollars to spend on the music. Was there a conscious choice to go with more recognisable stuff than the first game, or is it just a by-product of the game being set 10/20 years on?Īlex Cox: We've spent ages on the tracklist. RPS: I love the soundtrack - I was just haring around 1950s New York to Buddy Holly's That'll Be The Day. Read on for the chatty chap's thoughts on the game's excellent music, open world versus linear and whether 'dickcheese' is an authentic 1950s swear word or not. ![]() So I decided to ask him about the game instead. While I was there, I made Alex Cox, one of the game's producers, an offer he couldn't refuse. ![]() Last week, I finally got to spend a little quality time with the rarely-seen, much-delayed Mafia 2. ![]() While the former stands very close to being called a fully integrated IDE, the latter features a test-driven environment. What are the features of Visual Studio Code? As soon as you get familiar with the software, you can easily stage files, make commits, and review diffs directly from the editor. It also makes working with SCM providers a lot easier than before. In addition to this, VS Code goes beyond highlighting syntax by autocompleting code using function definitions, variable types, and imported modules. Oh, and did we mention a debugger for JavaScript? Git support? Diffs? Easy integration with tools like Make, Ant, Gulp, Jake, Rake, MSBuild? It can validate JavaScript files according to your preferred rules, preview Markdown files, show colour preview annotations for CSS, LESS and SASS, and gives you IntelliSense, intelligent refactoring, "find all references" and more. Open a C# project.json file and it'll load that project plus reference projects open a *.sln and it'll load the referenced *.csproj's and any sibling or descendant project.json-files. So, they're currently running an oil consumption test on it. My wife took it to the dealership a couple of weeks ago when the engine light was on and they told her that it was completely out of oil, but it wasn't even time for an oil change yet. And when it happens and we turn the engine off, it shuts down rough - like an old diesel engine. The engine usually doesn't run rough while driving, only while idling. So, they use the computer to manually turn off the check engine light. When we take it to the dealership, they hook up the computer to diagnose the problem and it won't even produce a code to tall them what the problem is. Recently though, within the last couple of months, the check engine light has been coming on too. My wife says that the engine has been rough since we bought the car in 2010, but it didn't do it very frequently until about six months ago. Not to mention all the other issues like replacing my transmission at 90000km ($5000), replacing throttle body ($500) because my car would just stall while driving in the middle of the road as well as replacing my muffler. Not until 2-3 months later until I noticed a rather loud ticking and clanking noise coming from my engine I took the car into the shop for an oil change being that I have driven 50-100 km past the suggested kilometers to do an oil change to find out THERE WAS NO OIL!!!!! I have owned several vehicles that Ill drive 1000 km+ past the oil change date and MAYBE the engine would be half a quart low AT THE MOST!!! So now I find myself getting oil changes every 2000km if not adding oil which is F* ridiculous. I purchased my 2010 Chevy Equinox used with 65000km in winter 2015, stylish and very comfortable vehicle. ![]() From a creative standpoint, the contrast of barren gunk zones transforming into flourishing havens were fun and inspiring for the team to work on.
